RESUMO

Due to the variability of body coloration and morphological similarity among closely related species, unresolved issues and debates still persist in the taxonomic study of the genus Sycanus from China. In this study, we conducted phylogenetic analyses and species delimitation for Sycanus in China based on a COI DNA barcoding dataset comprising 81 samples. The results revealed that all the samples could be classified into 12 species by integrating molecular analyses with morphological comparison. This paper provides a comprehensive systematic review of the Sycanus species found in China, including descriptions of three new species: S. taiwanensis Zhao & Cai sp. nov., S. flavicorius Li & Cai sp. nov., and S. hainanensis Wang & Cai sp. nov. Furthermore, it is proposed that S. croceovittatus Dohrn, 1859, S. leucomesus Walker, 1873, and S. villicus Stål, 1863, are three synonyms of S. bifidus (Fabricius, 1787); S. bicolor Hsiao, 1979, is a synonym of S. versicolor Dohrn, 1859; and S. hsiaoi Maldonado-Capriles, 1990, is a synonym of S. marginellus Putshkov, 1987. Additionally, brief biological information is provided for two species, S. falleni Stål, 1863, and S. croceus Hsiao, 1979.

Interleukin-36 (IL-36) signaling plays an important role in promoting CD8+ T cell-mediated antitumor immune responses. The role of IL-36 signaling in CD8+ T cells that are involved in host immune responses during human immunodeficiency virus-1 (HIV-1) infection has not been characterized. Sixty-one patients living with chronic HIV-1 infection and 23 controls were enrolled in this study. The levels of IL-36 cytokine family members were measured by enzyme-linked immunosorbent assay. Purified CD8+ T cells were stimulated with recombinant IL-36gamma (1 or 10 ng/mL). The expression of inhibitory receptors, the secretion of cytotoxic molecules and interferon-gamma, and the mRNA levels of apoptosis-related ligands were assessed to evaluate the effect of IL-36gamma on CD8+ T cell function in vitro. There were no significant differences in IL-36alpha, IL-36beta, or IL-36 receptor antagonist levels between patients living with chronic HIV-1 infection and controls. Plasma IL-36gamma levels were reduced in patients living with chronic HIV-1 infection. Perforin, granzyme B, and granulysin secretion, as well as tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) and Fas ligand (FasL) mRNA expression, but not programmed death-1 (PD-1) or cytotoxic T lymphocyte-associated protein-4 (CTLA-4) expression was downregulated in CD8+ T cells from patients living with chronic HIV-1 infection. The addition of both 1 and 10 ng/mL IL-36gamma enhanced perforin, granzyme B, granulysin, and interferon-gamma secretion by CD8+ T cells without affecting PD-1/CTLA-4 or TRAIL/FasL mRNA expression in CD8+ T cells from patients living with chronic HIV-1 infection. The addition of 1 ng/mL IL-36gamma also promoted perforin and granzyme B secretion by HIV-1-specific CD8+ T cells from patients living with chronic HIV-1 infection. The reduced IL-36gamma levels in patients living with chronic HIV-1 infection might be insufficient for the activation of CD8+ T cells, leading to CD8+ T cell exhaustion.

As one of the most significant pathological changes of diabetic nephropathy (DN), tubulointerstitial fibrosis (TIF) had a close relationship with tubulointerstitial inflammation (TI), and the occurrence of TI could have resulted from the disrupted tight junctions (TJs) of renal tubular epithelial cells (RTECs). Studies have demonstrated that sodium butyrate (NaB), a typical short chain fatty acid (SCFA), played an important regulatory role in intestinal TJs and inflammation. In this study, our in vivo and in vitro results showed that accompanied by TI, renal tubular TJs were gradually disrupted in the process of DN-related TIF. In HG and LPS co-cultured HK-2 cells and db/db mice, NaB treatment regained the TJs of RTECs via the sphingosine 1-phosphate receptor-1 (S1PR1)/AMPK signaling pathway, relieving inflammation. Small interfering RNA of S1PR1, S1PR1 antagonist W146 and agonist SEW2871, and AMPK agonist AICAR were all used to further confirm the essential role of the S1PR1/AMPK signaling pathway in NaB's TJ protection in RTECs in vitro. Finally, NaB administration not only improved the renal function and TIF, but also relieved the TI of db/db mice. These findings suggested that the use of NaB might be a potential adjuvant treatment strategy for DN-associated TIF, and this protective effect was linked to the TJ modulation of RTECs via the S1PR1/AMPK signaling pathway, leading to the improvement of TI.

AIMS AND OBJECTIVES: This study explores the situation of workload, work-family conflict and job burnout among primary health workers in China in the context of COVID-19 and identifies the mediating effect of work-family conflict between workload and job burnout. BACKGROUND: Since the breakout of the COVID-19 pandemic, primary health workers have been working on the frontline of the epidemic and may experience increasing workload, work-family conflict and job burnout. It is important to focus on the issue of how to alleviate job burnout of primary health workers. DESIGN: A cross-sectional study (STROBE) was used. METHODS: Data were collected from 785 primary health workers in China. Multiple regression analysis was used to examine the mediating effect of work-family conflict between workload and job burnout. RESULTS: 18.7%, 10.4% and 39.5% of respondents had high job burnout in the dimensions of emotional exhaustion, depersonalization and personal accomplishment, respectively. 34.6% of the respondents had high or very high workload, and 12.8% of the respondents had high or very high work-family conflict. Results of multiple regression analysis indicated that work-family conflict mediated the relationship between workload and job burnout. Workload (ß = .163, CI = .207-.549) and work-family conflict (ß = .211, CI = .311-.640) positively influenced job burnout, and workload (ß = .428, CI = .375-.508) positively influenced work-family conflict. CONCLUSION: The study indicated that primary health workers experienced a high level of job burnout, especially in the personal accomplishment dimension. Furthermore, this study verified the mediating effect of work-family conflict between workload and job burnout. RELEVANCE TO CLINICAL PRACTICE: Some interventions for alleviating workload, work-family conflict and job burnout should be taken, including workplace assistance programmes, family-friendly policies and a well-integrated healthcare system. NO PATIENT OR PUBLIC CONTRIBUTION: This study does not involve patient or public contribution in any part. IMPACT STATEMENT: Nurses and other primary health workers are health gatekeepers of residents and play a vital role in the healthcare system. Due to the breakout of COVID-19, they have taken more work and are more vulnerable to work overload, work-family conflict and the consequent job burnout. Some interventions should be taken to effectively alleviate their job burnout and improve their health and performance.

AIMS: FoxO1 is an important factor in the ß-cell differentiation in type 2 diabetes mellitus (T2DM). Sirt3 is found to be involved in FoxO1 function. This study investigated the role of Sirt3 in the ß-cell dedifferentiation and its mechanism. METHODS: Twelve-week-old db/db mice and INS1 cells transfected with Sirt3-specific short hairpin RNA (shSirt3) were used to evaluate the dedifferentiation of ß-cell. Insulin levels were measured by enzyme linked immunosorbent assay. The proteins of Sirt3, T-FoxO1, Ac-FoxO1 and differentiation indexes such as NGN3, OCT4, MAFA were determined by western blot or immunofluorescence staining. The combination of Sirt3 and FoxO1 was determined by the co-immunoprecipitation assay. The transcriptional activity of FoxO1 was detected by dual luciferase reporter assay. RESULTS: Both the in vivo and in vitro results showed that Sirt3 was decreased along with ß-cell dedifferentiation and decreased function of insulin secretion under high glucose conditions. When Sirt3 was knocked down in INS1 cells, increased ß-cell dedifferentiation and lowered insulin secretion were observed. This effect was closely related to the amount loss and the decreased deacetylation of FoxO1, which resulted in a reduction in transcriptional activity. CONCLUSION: Downregulation of Sirt3 contributes to ß-cell dedifferentiation in high glucose via FoxO1. Intervention of Sirt3 may be an effective approach to prevent ß-cell failure in T2DM.

Species in Ectrichodiinae are known for their prey specialization on millipedes. However, knowledge of the morphological adaptations to this unique feeding habit was limited. In the current study, we examined the microstructures of the antennae, mouthparts, and legs of four millipede feeding ectrichodiines, Ectrychotes andreae (Thunberg, 1888), Haematoloecha limbata Miller, 1953, Labidocoris pectoralis (Stål, 1863), and Neozirta eidmanni (Taueber, 1930), and compared them with those of three species of tribelocephalines, a group closely related to Ectrichodiinae. On the antennae, we found four types of antennal sensilla. On the mouthparts, we recognized four types of labial sensilla. Sampled ectrichodiines have distinctly more and denser slightly transverse ridges on the external side of mandibles than tribelocephalines. E. andreae and H. limbata possess numerous small papillae fringed with densely arranged finger-print-like grains on the trochanter and femur; these probably facilitate the immobilization of prey. Overall, our study illustrates, at a microstructural level, the remarkable morphological adaption of prey manipulation in ectrichodiine, and has enhanced our understanding about stenophagy in the family Reduviidae.

Objective:To investigate the application value of near-infrared autofluorescence imaging in identifying and protecting parathyroid glands in endoscopic thyroid surgery. Methods:From May 2022 to February 2023, 158 patients who underwent endoscopic thyroid surgery in the Department of Thyroid and Breast Vascular Surgery of Guilin People's Hospital were selected. The endoscopic fluorescence camera system was used to monitor the parathyroid glands under autofluorescence during endoscopic thyroid surgery. A total of 214 pieces were collected, among which the first 15 cases that could not be preserved in situ during the operation needed to be autotransplanted or the tissue clamped parts that could not be clearly identified as parathyroid glands were sent to fast-frozen pathology to determine whether they were parathyroid glands. Results:Among the first 15 patients who could not be preserved in situ during the operation or whose anatomy could not be clearly defined, 23 parathyroid glands were detected by autofluorescence imaging, 21 parathyroid glands were confirmed by pathology, and 2 were adipose tissue, with an accuracy rate of 91.30%; 158 patients underwent surgery Blood calcium decreased 2 hours after operation compared with preoperative blood calcium(P<0.05), decreased blood calcium 5 days after operation compared with preoperative blood calcium(P<0.01), and increased slightly 5 days after the operation compared to blood calcium 2 hours after the operation, but the difference was not statistically significant(P>0.05); while comparing parathyroid hormone(PTH), PTH at 2 hours after operation decreased significantly compared with PTH before operation(P<0.01), and PTH at 5 days after operation compared with PTH before operation PTH also decreased(P<0.01), but increased compared with PTH 2 hours after operation(P=0.001). Conclusion:In laparoscopic thyroid surgery, the application of near-infrared autofluorescence imaging technology can help surgeons quickly identify and protect parathyroid glands, and reduce the incidence of permanent hypoparathyroidism. Combining autofluorescence imaging, visual anatomy recognition under magnification of laparoscope, and intraoperative frozen pathological examination "trinity" method can improve the success rate of parathyroid gland recognition.

The spider mites Eotetranychus sexmaculatus, Eutetranychus orientalis and Oligonychus biharensisin are severe pests of rubber trees in China. The predatory mite Neoseiulus barkeri has been found to be a natural enemy of these three pests, while nothing is known about the biological performance of this phytoseiid predator against these phytophagous mites. In this study, the development, survivorship, reproduction, adult longevity, fecundity, sex ratio and population growth parameters of N. barkeri fed on these pests were evaluated in comparison to the factitious prey Tyrophagus putrescentiae in the laboratory at 25 ± 1 °C, 75 ± 5% relative humidity and a 12:12 (L:D) h photoperiod. The results showed that N. barkeri could develop from egg to adult and reproduced successfully on the three preys. The survival rate of N. barkeri from egg to adult was higher when fed on E. orientalis (100%) and T. putrescentiae (100%) than when fed on O. biharensisin (93.60%) and E. sexmaculatus (71.42%). The shortest and longest generation time for N. barkeri were observed on E. orientalis with 6.67 d and E. sexmaculatus with 12.50 d, respectively. The maximum fecundity (29.35 eggs per female) and highest intrinsic rate of increase (rm = 0.226) were recorded when N. barkeri fed on E. orientalis, while feeding on E. sexmaculatus gave the minimum fecundity (1.87 eggs per female) and lowest reproduction rate (rm = 0.041). The values of these parameters for N. barkeri evaluated on O. biharensisin were found to be comparable to those obtained on T. putrescentiae. The sex ratio of N. barkeri progeny on the preys mentioned above, apart from O. biharensisin, was female biased. According to the findings, N. barkeri could serve as a promising biocontrol agent against E. orientalis and O. biharensisin, and possibly E. sexmaculatus on rubber trees.

HIV-infected individuals are susceptible to Mycobacterium tuberculosis (M.tb) infection and are at high risk of developing active tuberculosis (TB). Interferon-gamma release assays (IGRAs) are auxiliary tools in the diagnosis of TB. However, the performance of IGRAs in HIV-infected individuals is suboptimal, which limits clinical application. Interferon-inducible protein 10 (IP-10) is an alternative biomarker for identifying M.tb infection due to its high expression after stimulation with M.tb antigens. However, whether IP-10 mRNA constitutes a target for the diagnosis of TB in HIV-infected individuals is unknown. Thus, we prospectively enrolled HIV-infected patients with suspected active TB from five hospitals between May 2021 and May 2022, and performed the IGRA test (QFT-GIT) alongside the IP-10 mRNA release assay on peripheral blood. Of the 216 participants, 152 TB patients and 48 non-TB patients with a conclusive diagnosis were included in the final analysis. The number of indeterminate results of IP-10 mRNA release assay (13/200, 6.5%) was significantly lower than that of the QFT-GIT test (42/200, 21.0%) (P = 0.000026). IP-10 mRNA release assay had a sensitivity of 65.3% (95%CI 55.9% - 73.8%) and a specificity of 74.2% (95%CI 55.4% - 88.1%), respectively; while the QFT-GIT test had a sensitivity of 43.2% (95%CI 34.1% - 52.7%) and a specificity of 87.1% (95%CI 70.2% - 96.4%), respectively. The sensitivity of the IP-10 mRNA release assay was significantly higher than that of QFT-GIT test (P = 0.00062), while no significant difference was detected between the specificities of these two tests (P = 0.198). The IP-10 mRNA release assay showed a lower dependence on CD4+ T cells than that of QFT-GIT test. This was evidenced by the fact that the QFT-GIT test had a higher number of indeterminate results and a lower sensitivity when the CD4+ T cells counts were decreased (P < 0.05), while no significant difference in the number of indeterminate results and sensitivity were observed for the IP-10 mRNA release assay among HIV-infected individuals with varied CD4+T cells counts (P > 0.05). Therefore, our study suggested that M.tb specific IP-10 mRNA is a better biomarker for diagnosis of TB in HIV-infected individuals.

Microbial-induced carbonate precipitation (MICP) is being investigated to repair concrete cracks because of its good durability and compatibility with cementitious matrix. However, during the in-situ application, the repairing often lasts weeks, even months. And the strength regain is quite low. The repairing time is largely determined by the CaCO3 yield, and the strength regain after the repair is closely related to the cohesion and bonding strength of CaCO3 itself. Thus, the purpose of this paper is to obtain an efficient precipitation of bio-CaCO3 with both high yield and good cohesion to improve the in-situ repairing efficiency. Firstly, the most influential factors on urease activity were screened and the precipitation kinetics were detailly investigated. The results show that the CaCO3 with the largest yield and cohesion was obtained when the bacterial concentration was 107 cells/mL and the concentration of urea and calcium was both 0.5 M at 20 °C. This weight loss of bio-CaCO3 was 9.24% under ultrasonic attack. Secondly, two models were established to quantify or semi-quantify the relationship between the most influential factors and the yield and cohesion of precipitates, respectively. The results showed the order of contribution for bio-CaCO3 precipitation was calcium ions concentration > bacterial concentration > urea concentration > temperature > initial pH. According to these models, the required yield and cohesion of CaCO3 by engineering could be obtained by adjusting affecting factors. Models were proposed for guiding the application of MICP in practical engineering. KEY POINTS: • Screened the most affecting factors on urease activity and investigated the precipitation kinetics. • Obtained optimal conditions of bio-CaCO. • Established two models in order to give some guidance for practical civil engineering.

A unified model based on the ultrasonic lag phase slope is developed for measuring the lubricating film thickness at a large scale. The ultrasonic lag phase of adjacent waves instead of the phase of overlapped waves is calculated as a function of the ultrasonic frequency and film thickness. The slope of the ultrasonic lag phase is determined correspondingly, which is linearly proportional to the lubricating film thickness. Both the finite element analysis and tests on the lubricating film thickness are performed to verify the proposed method. The results show that despite the fluctuations of the lag phase, the lag phase slope can be used for measuring the lubricating film thickness at a large scale from 0.1 µm to 170 µm.

Scutellista ciruela Fonscolombe has a significant controlling effect on the rubber tree pest, Parasaissetia nigra Nietner. To identify the optimal conditions for the population growth of S. caerulea, we assessed how temperature, photoperiod, and supplementary nutrition affected its development and reproduction. The results demonstrated that the number of eggs laid and parasitism rates of S. caerulea were the highest at 33 °C. The developmental rate of S. caerulea was the fastest and the number of emerged adults the highest. The number of eggs laid and the parasitism rates increased when the light duration increased within a day. Females did not lay any eggs when the whole day was dark. At a photoperiod of 14:10 (L:D), the developmental duration was the shortest and the number of emerged adults was the highest. Adult life span was the longest under a 12:12 (L:D) photoperiod. During the adult stage, supplementary nutrition, such as sucrose, fructose, honey, and glucose, increased the life span of S. caerulea. The life span of S. caerulea was longer when provided with a supplementary diet of sucrose or honey, compared to other tested diets. The results suggested that the most suitable conditions for S. caerulea's population growth were the following: 30 to 33 °C, with 12 to 14 h of daylight, and the provision of sucrose or honey as supplemental diet for the adults.

ABSTRACT: The association between age and mental health symptoms among persons with HIV (PWH) is inconsistent, and little is known about the mediators and moderators of this association. This study aimed to examine the association between age and mental health symptoms, as well as the mediators and moderators of perceived discrimination. Data were from 1,304 PWH who completed a cross-sectional survey in five areas of China. Multiple linear regressions showed that younger age was significantly associated with more severe mental health symptoms and that perceived discrimination moderated this relationship. The Sobel test showed that perceived discrimination also mediated the association between age and mental health symptoms. Our study indicates that perceived discrimination shapes the association between age and mental health symptoms among PWH and highlights the importance of designing age-tailored mental health intervention strategies for perceived discrimination among young PWH. Interventions addressing discrimination are necessary to help improve mental health, especially for young PWH.

The recycled concrete aggregates have high porosity and water absorption, which hinders their utilization in concrete production. Microbial-induced calcium carbonate precipitation was regarded as a very promising method for strengthening recycled aggregates. However, the uneven distribution of CaCO3 on surface of aggregates encountered in the current bio-deposition treatment weakened the efficiency, especially in the aspect of the decrease of water absorption. Therefore, this study innovatively applied a sodium alginate aided bio-deposition treatment to improve the uniform distribution of biogenic CaCO3. The principle was that sodium alginate was used to uniformly "fix" the bio-agents (urea or bacterial cells) on the surface of recycled aggregates, which was supposed to promote the uniform in-situ precipitation of CaCO3 on the surface of aggregates, and hence effectively blocking surface pores, and reducing the water absorption of the aggregates. Two concentrations of sodium alginate (0.2w% and 0.5w%) and four sodium alginate aided bio-deposition treatments were studied. It was found that CaCO3 (a mass increase of 4.05%) was formed on the aggregates after the suitable sodium alginate aided bio-deposition treatment. The participation of sodium alginate made CaCO3 uniformly deposited on full surface of the aggregates, resulting in a significant decrease (42.10%) of water absorption. The biogenic CaCO3 showed limited mass loss under ultrasonic attack, indicated a strong cohesion and bonding strength with aggregates. The results demonstrated that sodium alginate-aided bio-deposition treatment can enhance the efficiency, which was beneficial to improve the quality of recycled aggregates and their utilization of recycled aggregates in concrete production. OBJECTIVES: This study aimed to examine the trajectories of depressive symptoms over a 5-year period in a sample of community-dwelling older adults with disabilities in rural China, and the association between duration of instrumental support primarily provided by adult children and the trajectories of depressive symptoms. METHODS: Data were drawn from three waves of the China Health and Retirement Longitudinal Study (2011-2015). The sample included 1,466 older adults living in rural areas aged 60 and over who had at least one child in all three waves. Duration of instrumental support provided by adult children was measured by the number of waves that older adults received instrumental support primarily from any adult children in the data collection. Growth mixture modelling was used to identify the trajectory classes of depressive symptoms among these older adults, and logistic regression was used to examine the association between duration of instrumental support primarily provided by adult children and the trajectory classes of depressive symptoms. RESULTS: This study identified two distinct trajectories of depressive symptoms among the respondents: high but decreasing levels of symptoms and persistently low levels of symptoms. Respondents who received a longer duration of instrumental support primarily from adult children were more likely to have high but decreasing symptoms after controlling for other covariates. CONCLUSION: The findings have important implications for promoting the mental health of older adults with disabilities in rural China. There are still very limited long-term care services and programs are available for older adults, particularly in rural China. It is critical to develop these services at both community and institutional levels to meet the care needs of older adults and their families.

BACKGROUND: Renal interstitial fibrosis (RIF) is one of the main features of diabetic nephropathy (DN), but the molecular mechanisms mediating RIF in DN has yet been fully understood. S100A8 and S100A9 are the proteins associated with immune and inflammation response. Here we reported the expression of S100A8 and S100A9 were significantly increased on tubular epithelial cells in diabetic kidneys through a proteomic analysis. METHODS: We detected the expression of S100A8/A9 in diabetic kidneys by using immunoblotting, real-time PCR and immunostaining. RNA silencing and overexpression were performed by using S100A8/A9 expression/knockdown lentivirus to investigate the connection between S100A8/A9 and epithelial to mesenchymal transition (EMT) process. We also identify the expression of TLR4/NFκB pathway-related molecules in the case mentioned above. Afterwards a CO-IP assay was used to verify that compound AB38b ameliorates the EMT by interfering S100A8/A9 expression. RESULTS: The expression of S100A8 and S100A9 were significantly increased on tubular epithelial cells in diabetic kidneys. S100A8/A9 knocking-down alleviate and over-expression promote the renal interstitial fibrosis of diabetic mice. Mechanically, high levels of S100A8/A9 expression in tubular epithelial cells during diabetic condition activated the TLR4/NF-κB signal pathway which promoted the EMT process and finally led to RIF progression. S100A8/A9 knockdown ameliorated RIF of diabetic mice. Further experiments revealed that compound AB38b inhibited the EMT progression of tubular epithelial cells induced by S100A8/A9 through interfering the expressions of S100A8/A9. CONCLUSIONS: Our study suggest that abnormal expression of S100A8/A9 in the disease condition promotes EMT process and RIF through TLR4/NF-κB signal pathway. Using small molecular inhibitor AB38b to inhibit the abnormal expressions of S100A8/A9 might be a novel therapeutic strategy in treating DN.

PMMA bone cement has been clinically used for decades in vertebroplasty due to its high mechanical strength and satisfactory injectability. However, the interface between bone and PMMA is fragile and more prone to refracture in situ because PMMA lacks a proper biological response from the host bone with minimal bone integration and dense fibrous tissue formation. Here, we modified PMMA by incoporating borosilicate glass (BSG) with a dual glass network of [BO3] and [SiO4], which spontaneously modulates immunity and osteogenesis. In particular, the BSG modified PMMA bone cement (abbreviated as BSG/PMMA cement) provided an alkaline microenvironment that spontaneously balanced the activities between osteoclasts and osteoblasts. Furthermore, the trace elements released from the BSGs enhanced the osteogenesis to strengthen the interface between the host bone and the implant. This study shows the first clinical case after implantation of BSG/PMMA for three months using the dual-energy CT, which found apatite nucleation around PMMA instead of fibrous tissues, indicating the biological interface was formed. Therefore, BSG/PMMA is promising as a biomaterial in vertebroplasty, overcoming the drawback of PMMA by improving the biological response from the host bone.

The investigations of Au-bearing alloy materials have been of broad research interest as their relevant features exhibit significant advantages compared with pure Au. Here, we extensively investigate the compression behaviors of BaAu compounds via first-principles calculations and find that a high-pressure cubic phase is calculated to be stable above 12 GPa. Further electronic calculations indicate that despite the low electronegativity of Ba, Fd-3m-structured BaAu exhibits metallic characteristics, which is different from those of semiconducting alkali metal aurides that possess slight characteristics of an ionic compound. These findings provide a step toward a further understanding of the electronic properties of BaAu compounds and provide key insight for exploring the other Au-bearing alloy materials under extreme conditions.
